Nope. If memory serves, the only reason for that tag is to tell the CLR that it needs the full 4.0 framework, as opposed to the "Client Profile".
 
Since 4.5 is an in-place replacement for 4.0, telling it "4.0" really means "Whatever 4.x version is installed"
