You can try to add under line into content page.
 
<ajaxtoolkit:toolkitscriptmanager runat="server" enablescriptglobalization="true" xmlns:ajaxtoolkit="#unknown">
EnableScriptLocalization="true" ID="ScriptManager1" ScriptMode="Debug" CombineScripts="false" />

Solution 2

Solution : Drag a new toolkitscriptmanager on to the form and use any new ajax control on that page. This will work. Thumbs Up | :thumbsup:

Solution 3

Add Ajax Toolscript Manager on that page.
 
eg:
 
<%@ Register Assembly="AjaxControlToolkit" Namespace="AjaxControlToolkit" TagPrefix="cc1" %>
 
after register assebly, add toolscript manager:
 
<cc1:ToolkitScriptManager ID="ToolkitScriptManager1" runat="server">
      </cc1:ToolkitScriptManager>
