can you please help me?

What I have tried:

builder.Services.AddAuthentication(JwtBearerDefaults.AuthenticationScheme)
    .AddJwtBearer(options =>
    {
        options.TokenValidationParameters = new TokenValidationParameters
        {
            ValidateIssuer = true,
            ValidateAudience = true,
            ValidateLifetime = true,
            ValidateIssuerSigningKey = true,
            ValidIssuer = Configuration["JWT:Issuer"],
            ValidAudience = Configuration["JWT:Audience"],
            IssuerSigningKey = new SymmetricSecurityKey(Encoding.UTF8.GetBytes(Configuration["JWT:Key"]))

        };

    });

At a guess, you're missing the Configuration property from your Startup class:
C#
public class Startup
{
    public Startup(IConfiguration configuration)
    {
        Configuration = configuration;
    }

    public IConfiguration Configuration { get; }

    ...

use builder.Configuration["Jwt:xxxxxx"] instead of Configuration["Jwt:xxxxxx"] if you're using program.cs file.
