I've been tasked with my first commerical website, done a few intranet website before where I know the screen resolutions etc.

But what is the best size to work to when screen resolution is unknown?

Consider typical laptop screen sizes, and 1024 is probably the best "minimum" to assume. Beyond that, make your layout as fluid as possible and don't hard-code font sizes. You'll probably also have to consider mobile devices as well, so you'll need either a mobile version of the site, or work that into the main sight. If you want iPhone compatibility, you probably have to avoid using Flash and Silverlight for rich content.

which is a minimum resolution which every one should have minimum.
