If this is truly ASP.NET then yoiu can't - the Pen drive would be connected to the Client PC and there is no way the Server can detect that it exists, much less has been plugged in, for security reasons.

If you mean to detect it in a .NET winforms app, then have a look at this article: Detecting USB Drive Removal in a C# Program[^] - it covers insertion and removal.
