If you mean a pendrive attached to your client computer (the one running the browser) then pretty much, you can't - security won't let you. If you could, so could malicious sites - and that would be a Bad Thing. The client can access it and upload a file from it to you, or you can send him a download which he saves onto the pen drive, but you can't specify the destination from your website.

There is an exception to this, but it requires IE, ActiveX enabled and the user to confirm that your ActiveX control is allowed to run - and this happens only in 0.0001% of users as it's all disabled by default to prevent malicious sites doing exactly what you are trying...

If you are talking about a pendrive attached to your webserver, then it's just another drive so provided you have the appropriate access permissions (not guaranteed) you just access it via the drive letter as a normal path.
