He was thinking that because it's a pun, meaning "the big feminine piss-take", which seems appropriate, as it appears to be a self-portrait.
There was some BS going around (allegedly invented by some moron in a university, who had to publish something to justify his salary) that said the painting was named for Lisa Giocondo, which, being BS, can be ignored.
I can't remember where the name "Mona Lisa" came from, but, AIRI, it wasn't Da Vinci.

In Fiorentine "Monna" is a diminutive of "Madonna", which translates roughly to "Madame". So it would be Madame Lisa. It is a recurring venetian joke to call it the Mona Lisa, or " 'l quadro de la mona" ("the painting of the <female genitalia="">")
