Sounds like you have set as Windows Authentication in your Web.Config file of ASP.NET application but have not configured your application in IIS or configured in IIS but not set it to use Windows Authentication.

Go into Directory Security of your website and confirm that your website is configured for it by checking the checkbox to use 'Integrated Windows authentication'
