We want to get the client desktop screen shot using web application. Can anybody please suggest me the way to do this.

I don't think it's possible. Any web application has very limited access to a client's system for obvious security reasons.

Well, why do you want it? You best bet would be to ask the client to upload the screenshot by giving them a file upload option.

from me i think is very clear that you can not capture anything from client machine's without his permissions so once they allowed to do so then you more some activities from him to do.

like take example of facebook
when user have to change profile picture then they have to pic it or upload it by flash player

Can this be done using by creating browser plugin or using silverlight or Flex or Java Applet
