Why App_Data/ASPNETDB.MDF gets created automatically when i use profiles?What data it store? can I create a different database for this?If yes, What is the procedure?

When you use profiles, the ASPNETDB.MDF file contains the user log in details.
You can use a different database, but then you must take over responsibility for logging in and out users correctly.

Thanks. I saw this while creating webparts in my application and it is in the appliocation folder itself. Can I move this .mdf file to the DB? Can you please tell the procedure for that.

Um - an MDF file is a DB? Specifically, an Access DB?
